Identification of Dominant Microbial Communities on the Foliage of Zimbabwean Tobacco Using a Novel Tobacco-containing Medium

In order to better identify the dominant microorganisms on the leaves of Zimbabwean tobacco, tobacco components at concentrations(volume fractions) of 0%,2.5%,5.0% and 10.0% were added to simulate the microbial growth environment on the surface of tobacco leaves, to screen the concentration of tobacco leaves suitable for microbial growth, to explore suitable tobacco culture medium preparation conditions, to analyze the differences in the composition of the microbial communities of tobacco leaves at different concentrations, to isolate and identify specific microorganisms.The results showed that more specific microbial communities could be isolated when the tobacco leaf culture medium was adjusted to pH 7.0 at concentrations of 2.5% and 5.0% after sterilization.Metagenomic sequencing technology was used to determine the community distribution of microorganisms on the surface of tobacco leaves in Zimbabwe.It was found that Bacillus had a high relative abundance in the culture medium at concentrations of 0%,2.5% and 5.0%,and after the addition of tobacco to the culture medium, Terribacillus became the dominant species.The genera with higher relative abundance on tobacco culture medium at concentrations of 2.5% and 5.0% included Piscibacillus,Amphibacillus,Margalitia,etc.Through isolation and sequencing, 15 kinds of dominant microorganisms on the surface of tobacco leaves were identified, such as Bacillus species of the genus Bacillus,Bacillus veleis and Bacillus subtilis.

Research Progress on Land Suitability Evaluation

Land suitability refers to the suitability of a particular type of land for a particular use. Global population growth and environmental problems pose serious challenges to land security. Currently, the uneven distribution of land resources in China affects agricultural production decisions and jeopardizes national food security and sustainable development. Land suitability evaluation is the key to ensuring the rational planning and utilization of land resources, which is of great significance in optimizing land production potential, alleviating human-land conflicts and achieving land sustainability. This paper systematically reviews research progress of land suitability evaluation on the connotation, spatial scale, indicator systems, methods and ideas by the literature review method, and points out its shortcomings and the direction of future research, with a view to providing reference for optimizing the structure of land resource utilization and achieving sustainable development goals. Future research should follow a path that includes learning from the theories of geography, ecology, economics and other disciplines to strengthen the basic theoretical research, focusing on ecological orientation, improving evaluation indicators, expanding application space of the research results, and innovating evaluation methods and models, etc.

Analysis of Yield Difference of Different Sesame Types at Different Harvest Periods

In order to clarify the difference of yield reduction causes of different sesame varieties after maturity.In this paper, we took four sesame varieties(three conventional varieties, one non-shattering variety) as experimental material to study.The sesames were harvested in four stages(not open, just open, half open, full open) according to the degree of capsule dehiscent after maturity.We compared and analyzed the yield traits of these two types of sesame at different harvest periods.The results showed that the seeds per capsule, 1 000-seed weight and the seed yield per plant of three conventional sesame varieties decreased significantly with the increase of the capsule dehiscent degree.1 000-seed weight were the seed yield per plant of non-shattering sesame variety decreased significantly with the increase of capsule dehiscent degree.The results showed that the seeds per capsule and 1 000-seed weight were significantly and positively correlated with the seed yield per plant of three conventional varieties, therefore the seeds per capsule and 1 000-seed weight were the main factors that caused the decrease of the seed yield per plant of conventional variety.There was a significantly positive correlation between 1 000-seed weight and the seed yield per plant of non-shattering variety, so 1 000-seed weight was the main factor that caused the decrease of the seed yield per plant of non-shattering variety.

Introduction Experiment of Barley in the Arid Region of Central Gansu Province

[Objective] To screen new barley varieties suitable for planting in the arid areas of central Gansu Province.[ Method] Seven barley strains(species) were introduced from Gansu Academy of Agricultural Sciences in 2021-2022 for screening experiments in Dingxi City.[Result]The results showed that the protein content of all tested strains(species) was relatively high, ranging from 14.2% to 16.1%;except for 1008-1,which is higher, the fullness of >2.5 mm is lower than 70% for all others; the thousand grain weight is below 40.1 g; in terms of production, Y2017 has an equivalent yield of 2 950.43 kg/hm~2,which is 17.2% higher than the control Ganpi 7,ranking first among the tested strains(species).The other tested strains(species) have all reduced production compared to the control Ganpi 7.[Conclusion]The comprehensive analysis results indicate that Y2017 has good comprehensive traits and is suitable for demonstration and promotion planting of feed barley in the arid areas of central Gansu Province.

Study on Adaptability of Different Flue-cured Tobacco Varieties (Line) in Tobacco-growing Area of Luonan

To identify new flue-cured tobacco lines suitable for cultivation in the Luonan tobacco-growing area, a field experiment was carried out in Luonan County.Using Yunyan 99 and K326 as control varieties, the adaptability of new tobacco varieties such as 9K57,9K30,9L51,Y87NS1,HN230,CF240(DX06),HNCS04,and 9K54 were studied.The evaluation covered aspects including the growth period, botanical traits, main agronomic traits, disease resistance, raw tobacco appearance quality, and key economic traits.The results showed that 9K30 exhibited good comprehensive characteristics, recommending further promotion in the Luonan area.

Study on Morphological Differences and Biomass Statistics of Melastoma dodecandrum Under Different Cultivation Substrates

In order to explore the effects of different cultivation substrates on the growth of Melastoma dodecandrum,the most suitable substrate ratio for Melastoma dodecandrum growth was selected,providing a theoretical and material basis for the promotion and application of Melastoma dodecandrum in gardens;using Melastoma dodecandrum as the experimental material,peat soil,fine vermiculite,coarse vermiculite,volcanic stone,and perlite were used as cultivation substrates in different volume ratios.A total of 12 treatments were designed for pot experiments to determine the morphological indicators,relative chlorophyll content,and biomass indicators of Melastoma dodecandrum and conduct comprehensive evaluation and analysis.The results showed that treatment 1(peat soil∶ fine vermiculite=3∶ 1) and treatment 3(peat soil∶ fine vermiculite=1∶ 3) had the best growth status of Melastoma dodecandrum and could be used as a cultivation substrate for ground mulberry.

Visualized Analysis of Medicinal Dietary Plants of the Gastrodia elata at Home and Abroad Based on Bibliometrics

[Objective]In order to comprehensively understand the development and latest trends of Gastrodia elata, this paper analyzed this field at home and abroad based on bibliometrics, and revealed its research hotspots and difficulties. [Method]Using Web of Science(WoS) core collection database and China National Knowledge Network(CNKI) database as data sources, CiteSpace and VOSviewer were used to visualize the number of publications, authors, institutions and keyword co-occurrence networks. [Result]Gastrodia elata has a medicinal and edible history of more than two thousand years in China,Chinese scholars have made outstanding contributions in the field of Gastrodia elata. The research in this field reached its peak and then became stable in 2019. The focus of researches are the pharmacological effects of the active components, the therapeutic effects of its compound and the separation and quality evaluation of the chemical components of Gastrodia elata. With the development of the health industry, Gastrodia elata has entered the homologous catalogue of medicine and food, and the research and application of gastrodia will be diversified. [Conclusion]Through visual analysis of Gastrodia elata related literature, this study is helpful to grasp the research status of Gastrodia elata. Green planting and processing of Gastrodia elata and modern industry are the future development direction of this research field.

The Effect of Bamboo Sawdust Substrate Cultivation on Agronomic Traits and Nutritional Quality of Stropharia rugosoannulata

[Objective] To optimize the substrate formula for the cultivation of Stropharia rugosoannulata. [Method] Utilizing waste materials from three bamboo species(Phyllostachys edulis, Phyllostachys praecox, and Bambusa multiplex) including bamboo branches, leaves, tips, and residues from bamboo pole processing as cultivation materials for S. rugosoannulata, three bamboo sawdust ratios(68%, 78% and 88%) were set up. By observing mycelial growth and fruiting time, the agronomic traits and nutritional components of S. rugosoannulata were compared to study the effects of different substrate ratios on the growth and quality of the mushroom. [Result] Bamboo forests produce over 70% waste materials; the carbon-nitrogen ratio of bamboo sawdust ranges from 48.00 to 53.21, with P and Mg content ranging from 0.47 to 1.33 mg/g, and K and Ca content ranging from 1.66 to 3.38 mg/g, indicating a rich content suitable for the growth of S. rugosoannulata; under the cultivation conditions using B. multiplex sawdust, the content of crude protein(378.2 g/kg) and crude polysaccharides(186.0 g/kg) in S. rugosoannulata was the highest, with the largest proportion of high-quality mushrooms, reaching 44.75%. [Conclusion] Bamboo sawdust is suitable for the cultivation of S. rugosoannulata, with B. multiplex sawdust showing the best cultivation effects, effectively improving the nutrition and yield of the mushroom, and effectively extending the industrial chain of the bamboo forest ecosystem.

The Effects of Reduced Phosphorus Fertilizer Application on the Agronomic Traits and Yield Quality of Flue-cured Tobacco in Phosphorus-rich Soil

To explore the feasibility of reducing phosphorus fertilizer application in phosphorus-rich tobacco-growing soils,a field experiment was conducted with three different phosphorus fertilizer gradients(T1 25% reduction,T2 50% reduction,CK no reduction;same topdressing).The agronomic traits,economic traits,and various quality indicators of flue-cured tobacco under different treatments were comprehensively analyzed.The results showed that the effective leaf number of flue-cured tobacco with 25% and 50% phosphorus reduction increased slightly compared to CK,by 8.4% and 10.7% respectively;the stem content in the upper leaves of flue-cured tobacco with 25% and 50% phosphorus reduction was significantly lower than CK by 1.24% and 3.78%;the nicotine content in all leaf positions with 25% phosphorus reduction was significantly higher than CK.Phosphorus reduction improved the economic traits of flue-cured tobacco,with yields of 25% and 50% phosphorus reduction treatments increasing by 6.9% and 5.5% respectively compared to CK,and the output value increased by 7 630.60 and 5 525.37 yuan/hm~2.Phosphorus reduction had a certain promoting effect on the sensory quality of middle leaves of flue-cured tobacco,with 25% phosphorus reduction significantly improving various quality evaluation indicators compared to CK,and the total quality evaluation score increased by 2.65;50% phosphorus reduction showed no significant difference compared to CK,with slight increases in all indicators except aftertaste.In summary,the economic traits of flue-cured tobacco with 25% and 50% phosphorus reduction were improved to varying degrees,and a 25% reduction in phosphorus fertilizer further improved the quality of tobacco leaves.Therefore,it is preliminarily considered feasible to reduce phosphorus fertilizer application by 25% in phosphorus-rich tobacco-growing soils.

Poria cocos has a variety of pharmacological effects,two powerful active ingredients of Poria cocos are polysaccharides and triterpenes. Polysaccharides of Poria cocos have functions of antitumor activities,liver protective effect,acts as a diuretic,antisenility effects,anti-inflammatory,decrease blood fat,enhance immunity,sleep-inducing effect. Triterpenes of Poria cocos has function of antitumor activities,liver protective effect,antisenility effects,anti- inflammatory,enhancing immunity.

Research and Application Advances of Yeast Two-hybrid Technique

Yeast two-hybrid,which studies the interaction among proteins in yeast eukaryotic cells by using genetics method,is a very effective molecular biology technology.It is widely used in the fields such as proteomics,cellular signal transduction and functional genomics.And it has become one of important experiment means in the research field of molecular biology.And many valuable discoveries have been achieved using this system.With the development of molecular biology,based on yeast two-hybrid,many techniques such as yeast one-hybrid,reverse two-hybrid,three-hybrid,and extranuclear two-hybrid were set up to be applied in the research on the interaction between protein-protein,protein-DNA,protein-RNA,and protein-ligand.The research advances of yeast two-hybrid and its relevant technologies and application were summarized.It could be forecast that yeast two-hybrid and its derived techniques would play more and more important role in the researches of the functional genomics and proteomics.
