Research Progress on Key Techniques of Distant Hybridization Breeding in Tree Peony
WANG Xiao-hui;CAO Jian-kang;PANG Jing-jing;Distant hybridization breeding in peonies is a key approach to broadening the genetic background of peony germplasm resources and creating breakthrough new varieties.This paper provides a systematic overview of the key technologies involved in distant hybridization of peonies,including parent selection,manifestations and overcoming methods of hybridization barriers,as well as hybrid identification and cultivation.It highlights that current research still faces challenges such as unclear mechanisms of reproductive isolation,poor generalizability of techniques,and long breeding cycles.Additionally,the paper offers insights into future directions for distant hybridization in peonies,aiming to provide theoretical references for both research and practical applications in peony distant hybridization breeding.
Progress in the Extraction Technology of Phellinus linteus Polysaccharides and Its Pharmacological Activities
YE Jing-jing;WANG Xiang-jun;CAO Ning-ning;Phellinus igniarius polysaccharides(PIPs) exhibit diverse pharmacological activities such as antitumor effects and immunomodulation,holding broad application prospects in the fields of healthcare,pharmaceuticals,and functional foods.Its extraction technologies mainly include hot water extraction,ultrasonic-assisted extraction,microwave-assisted extraction,and enzymatic hydrolysis,etc.Different processes have their own advantages and disadvantages in terms of extraction rate,purity,operability,and cost.Although significant progress has been made in the research on the extraction and bioactive functions of PIPs,there are still problems such as immature extraction processes and unstable biological activities.This paper systematically elaborates on the characteristics and application effects of various extraction technologies,and summarizes the multiple pharmacological activities of PIPs as well as their related mechanisms of action,aiming to provide a reference for in-depth research on its pharmacological efficacy and the transformation of achievements into health products,pharmaceuticals,and other products.
Effects of Different Mechanical Transplanting Densities on Yield and Quality of Fragrant Rice Lilixiang
HUANG Jin-hua;YIN Chun-yuan;LIU He-mei;To clarify the optimal mechanical transplanting density for the fragrant rice cultivar Lilixiang,fully exploit its yield potential,and preserve its high-quality traits,a field experiment was conducted with two mechanical transplanting density treatments:D1(330 trays/hm2,row spacing×plant spacing=30 cm×18 cm) and D2(375 trays/hm2,row spacing×plant spacing=30 cm×16 cm).The effects of different mechanical transplanting densities on tillering dynamics,grain yield,and rice quality of Lilixiang were investigated.Results indicated that mechanical transplanting density significantly regulated the growth,yield,and quality of Lilixiang.In terms of tillering dynamics,D2 exhibited a faster tiller growth rate and higher effective tiller number before the peak tillering stage,but tiller decline was pronounced in the later period,with a reduction of 46.3%.In contrast,D1 maintained stable population growth and a gentle tiller decline,with only a 16.4% reduction.Regarding yield performance,although D1 had a lower effective panicle number than D2,its seed-setting rate and 1 000-grain weight were higher.Consequently,both the theoretical yield(9.2 t/hm2) and actual yield(7.9 t/hm2) of D1 were significantly higher than those of D2(8.0 t/hm2 and 7.6 t/hm2,respectively).For quality characteristics,D1 outperformed D2 in milled rice rate and eating quality score,while D2 had a slightly higher head rice rate.Notably,D1 showed significantly higher chalky kernel rate and chalkiness degree than D2.No significant differences were observed in protein content,moisture content,or amylose content between the two treatments,all of which met the quality standards for high-quality fragrant rice.Comprehensive analysis revealed that under the ecological conditions of northern Henan,the mechanical transplanting density of 330 trays/hm2 with a row spacing×plant spacing of 30 cm×18 cm for Lilixiang achieved a synergistic improvement in high yield and high quality,providing a technical basis for the large-scale cultivation of this cultivar.
Comprehensive Evaluation of Salt Tolerance in Different Sorghum Cultivars Under Salt Stress
CHEN Chao;TIAN Chang-jiu;YE Si-hong;[Objective]To explore the response mechanisms of different sorghum cultivars to salt stress during the germination stage,screen germplasm resources with excellent salt tolerance,and provide theoretical basis and material support for high-yield cultivation of sorghum in saline-alkali soils and breeding of salt-tolerant cultivars.[Method]10 sorghum cultivars were used as experimental materials.A 150 mmol/L NaCl solution was used to simulate the salt stress environment,with deionized water as the control.11 indicators were determined,such as germination potential,germination rate,germination index,vigor index,root-shoot ratio,fresh weight of sprout per plant,and dry weight of root per plant.After calculating the relative values of each indicator,comprehensive evaluation was performed through correlation analysis,principal component analysis,and membership function method,and salt tolerance grades were classified by cluster analysis.[Result]Salt stress had a significant impact on all indicators of sorghum during the germination stage.Except for the dry weight of individual roots,the other 10 indicators showed extremely significant differences among the treatments(P<0.01).The vigor index had the largest variation range(-75.2%),while the dry weight of individual roots had a relatively small variation range(26.1%).Correlation analysis showed that most indicators were significantly positively correlated;the correlation coefficient between germination potential and germination rate was higher(0.904),and the correlation coefficient between vigor index and individual shoot length reached 0.956.Principal component analysis extracted 3 comprehensive factors with a cumulative contribution rate of 88.589%,identifying germination rate and vigor index as the core evaluation indicators.Based on cluster analysis of comprehensive evaluation values(D values),the 10 materials were divided into 4 categories:highly salt-tolerant,salt-tolerant,moderately salt-tolerant,and salt-sensitive.Among them,cultivar 8(D=0.94) had the strongest salt tolerance,and cultivar 20(D=0.05) had the weakest.[Conclusion]There are significant genotypic differences in salt tolerance among the tested sorghum germplasms during the germination stage.The screened highly salt-tolerant cultivars can provide high-quality parental materials for sorghum cultivation in saline-alkali soils and genetic improvement.
Effect of Different Maturity Variety Seeding Dates on Autumn-sown Buckwheat Under Warming Climate
SU Cai-xia;LI Mei-yun;ZHANG Xu;[Objective]To explore the influence of seeding date of different maturity varieties on autumn-sown buckwheat under warmish climatic conditions and to screen the most suitable buckwheat varieties and optimal seeding date for promotion in the Taixing area.[Method]Three varieties with different maturity levels were used as experimental materials to analyze the effects of three seeding date on the growth duration,main agronomic traits,and yield of buckwheat under relatively warmish climatic conditions.[Result]With the onset of warmish climatic conditions,the seeding dates of various varieties were gradually postponed,and their growth durations were correspondingly shortened;the shortening was particularly pronounced in early-maturing varieties.However,the yield of early-maturing and medium-maturing varieties increased significantly following late sowing.[Conclusion]The commonly used late-maturing buckwheat variety—widely promoted in the Taixing area—can be replaced by late-sown early-maturing or medium-maturing varieties,thereby shortening the buckwheat growth period and facilitating flexible crop rotation.This substitution also increases buckwheat output,avoids frost damage,and achieves a synergistic effect of stable and high yield.

Dynamic Characteristics of TMV-GFP Infection in Nicotiana benthamiana and Transcriptome Analysis of Petiole Tissues
CHEN Guo-liang;WANG Hai-juan;LEI Yu-chen;To elucidate the dynamic mechanism of Tobacco mosaic virus(TMV) infection in Nicotiana benthamiana and the responsive characteristics of host genes, this study inoculated 4-5-leaf-stage N. benthamiana plants with TMV-GFP, monitored the early infection process, and analyzed gene expression changes in petiole tissues using transcriptomics. The results showed that TMV-GFP invaded the inoculated petiole at 4 days post-inoculation(dpi) and achieved systemic infection throughout the whole plant by 7 dpi, with viral accumulation in petiole tissues increasing significantly over time. Transcriptome analysis indicated a strong correlation between samples collected at 4 dpi(PT4) and 7 dpi(PT7), and a total of 2043 differentially expressed genes(DEGs) were identified, including 1299 up-regulated and 744 down-regulated ones. GO enrichment analysis showed these DEGs are involved in biological processes such as lipid metabolism and ribosome biogenesis, while KEGG pathway enrichment analysis indicated that the DEGs were enriched in pathways including plant-pathogen interaction and phenylpropanoid biosynthesis. RT-qPCR validation confirmed that the expression patterns of five candidate genes were consistent with the transcriptome results, verifying the reliability of the data. This study clarifies the pattern of TMV transmission and accumulation in N. benthamiana, identifies key responsive genes and pathways, and provides a theoretical basis for research on plant resistance mechanisms against TMV movement and the development of antiviral strategies.

Effects of Acute Cold Stress on Physiological Functions of Early-day Chicks
SHAO Long-yi;XIE Jia;WU Zhi-xian;To evaluate the effects of acute cold stress(ACS) on the physiological functions of early-stage chicks, 32 one-day-old Hy-Line Brown layer chicks were randomly assigned to four groups(8 chicks per group), namely the thermoneutral control group(TN group), the 4-hour ACS group(ACS/4 h group), the 8-hour ACS group(ACS/8 h group), and the 12-hour ACS group(ACS/12 h group). At 7 days of age, chicks were exposed to ACS(ambient temperature 12 ℃, relative humidity 60%) and samples were collected to assess body temperature, fecal characteristics, and serum biochemical indicators. The results showed that after 4, 8, and 12 hours of acute cold stress exposure at 12 ℃ and 60% relative humidity, multiple physiological function indices of 7-day-old chicks, including serum protein content, thermoregulation, intestinal function, liver and kidney function, pancreatic function, and acid-base balance, were negatively affected to varying degrees. The degree of impairment increased with prolonged exposure time. These findings indicate that acute cold stress treatment has a significant negative impact on the physiological functions of early-stage chicks, providing a reference for establishing related experimental models and guiding further research.

Effects of Humus Soil on Tobacco Seedling Cultivation and Root Development
ZHANG Wei;XUE Meng;CHEN Liangsheng;To investigate the effects of humus soil on the growth and development of tobacco seedlings,a floating tray seedling culture was conducted to study the influence of different proportions of humus soil in the substrate on the growth and root characteristics of tobacco seedlings.The results showed that the seedling emergence rate,plant height,and stem diameter increased first and then decreased with the increase of humus soil and the decrease of peat in the substrate.The highest seedling emergence rate was achieved with the substrate formula yunnan peat∶ humus soil∶ coconut coir∶ perlite=25∶ 40∶ 10∶ 25,which was 3.92% higher than that of the control.Compared with the control,plant height,root length,root fresh weight,and root dry weight increased by 3.80%,5.34%,7.85%,and 36.59%,respectively.Meanwhile,the total root length,total surface area,average diameter,number of root tips,total root length by orders,and total surface area by orders were all superior to those in the other treatments.This study clarifies the optimal proportion of humus soil in tobacco seedling substrates,and the effects of the growth and root characteristics of tobacco seedlings was evaluated,which provides a theoretical foundation for the application of humus soil in substrate formulations.

Establishment of Regeneration System and Polyploid Induction of Chrysanthemum morifolium cv. ‘Chuju’
CUI Lian-qi;JIANG Jia-rui;ZHANG Wan;[Objective]To address the problems of variety degradation, yield reduction and quality decline in Chrysanthemum morifolium ‘Chuju’ caused by long-term reliance on asexual propagation methods such as cutting and ramet division. [Method]An in vitro regeneration system was constructed using leaf explants of ‘Chuju’ in this experiment, and polyploid induction research was carried out by colchicine treatment. [Result]The optimal disinfection method for ‘Chuju’ explants was 75% ethanol for 30 seconds combined with 1% sodium hypochlorite for 10 minutes. The optimal callus induction medium was MS supplemented with 0.5 mg/L 6-BA and 0.5 mg/L 2,4-D, and the induction rate could reach 100%. The optimal callus proliferation medium was MS with 1.0 mg/L 6-BA and 0.3 mg/L NAA, and the proliferation coefficient reached 4.75. The optimal callus differentiation medium was MS containing 2.0 mg/L 6-BA and 1.0 mg/L NAA, with a differentiation efficiency of 56.67%. The optimal rooting medium was 1/2 MS added with 0.1 mg/L IBA, resulting in a rooting rate of 88.89% and an average root length of 7.76 cm. In polyploid induction, no successful induction was achieved when calli were treated with colchicine, while the treatment of regenerated seedling stem segments with 0.025% colchicine for 48 hours showed the best effect, with a variation rate of 3.89% and a tetraploid rate of 75.00%, and triploids, tetraploids and chimeras were successfully obtained. Morphological, cytological and flow cytometry identification revealed significant differences between polyploids and diploids in plant height, leaf morphology, chlorophyll content, stomatal number and length, as well as DNA content. [Conclusion]The established in vitro regeneration system and polyploid induction technology for ‘Chuju’ in this study provide support for variety improvement and high-quality industrial development of ‘Chuju’.

Research Progress on the Impact of Salinization Stress on Soil Microbial Communities Based on Bibliometrics
WU Jin-fa;HE Meng-yuan;DING Shi-jie;Soil salinization is a global environmental challenge.It not only causes soil degradation and reduced productivity but also threatens the sustainability of ecosystems by disrupting the structure and function of microbial communities.This study focuses on microorganisms in salinized soils,aiming to reveal the dynamic evolution patterns of the research landscape through bibliometric analysis.A multidimensional bibliometric analysis was conducted using CiteSpace software based on the Web of Science Core Collection database(2000-2024) to explore publication characteristics,national and institutional collaboration networks,research hotspots,and evolutionary trends in this field.Results show that publication output in the field of microorganisms in salinized soils has demonstrated an upward trend.In terms of publication volume,China,India,and the United States are the top three countries,with the Chinese Academy of Sciences emerging as the most active research institution.Frontiers in Microbiology,with the highest number of publications in this domain,stands out as the leading journal.Furthermore,research on soil microorganisms in salinized soils has extensive interdisciplinary links with other fields,including plant science,environmental science,microbiology,and soil science.Current research focuses on functional microbiomes,salinization stress-plant interactions,and these areas are expected to remain significant in future research.This study presents the developmental trajectory and research hotspots of microorganisms in saline-alkali soils since 2000 and provides a scientific basis and reference for advancements in related research fields.

Research Advances in Extraction, Isolation, Purification and Content Determination of Methods Flavonoids

LI Xian-kun et al(College of Horticulture and Gardening of Hainan University,Danzhou,Hannan 571737)Yeast two-hybrid,which studies the interaction among proteins in yeast eukaryotic cells by using genetics method,is a very effective molecular biology technology.It is widely used in the fields such as proteomics,cellular signal transduction and functional genomics.And it has become one of important experiment means in the research field of molecular biology.And many valuable discoveries have been achieved using this system.With the development of molecular biology,based on yeast two-hybrid,many techniques such as yeast one-hybrid,reverse two-hybrid,three-hybrid,and extranuclear two-hybrid were set up to be applied in the research on the interaction between protein-protein,protein-DNA,protein-RNA,and protein-ligand.The research advances of yeast two-hybrid and its relevant technologies and application were summarized.It could be forecast that yeast two-hybrid and its derived techniques would play more and more important role in the researches of the functional genomics and proteomics.
