Research Progress on Soil Microorganism of Xanthoceras sorbifolia Bunge
JI Yuan-yuan;Sharentuya;YANG Yue-wen;Key Laboratory of State Forestry and Grassland Administration on Sandy Land Biological Resources Conservation and Cultivation;Inner Mongolia Ordos Forest Ecosystem Nationol Observation and Research Station;Xanthoceras sorbifolia Bunge is drought-tolerant, salt-tolerant and barren-tolerant.It is a kind of biomass energy tree with economic value.Its growth and development are closely related to soil microorganisms.The biological characteristics of Xanthoceras sorbifolia Bunge and the importance of its cultivation were discussed and the effects of structure and function of soil microbial community on Xanthoceras sorbifolia Bunge growth, nutrient supply, biological control and other aspectswere deeply analyzed based on the current research status and methods of soil microorganisms.The dominant position of soil microorganisms in nutrient supply was clarified.And the screening of growth-promoting microorganisms and its specific mechanism, the soil physicochemical properties and regulation mechanism of cultivation management measures on soil microbial community were also discussed.Finally, looking forward the future research directions, amis to provide a scientific basis for optimizing cultivation strategies, improving soil efficiency and promoting the green development of Xanthoceras sorbifolia industry.
Advances in the Breeding and Landscape Applications of Rare Medicinal and Ornamental Plants Cremastra appendiculata (D.Don) Makino
DENG Yu-ye;XIANG Zheng;YANG Sheng-fei;Cremastra appendiculata(D. Don) Makino, as a perennial herbaceous plant in the Orchidaceae family, is one of the medicinal plants for the traditional Chinese medicine “Shancigu”. It has extremely high medicinal and ornamental values. However, factors such as long-term excavation, ecological damage, difficulty in pollination, and limitations in asexual reproduction have made Cremastra appendiculata(D. Don) Makino an endangered species, thereby limiting its large-scale application in landscaping. By analyzing its ecological habits and morphological characteristics, and drawing on the experience of commercial orchids in garden applications, propagation techniquess from aspects such as division propagation, seed propagation, and tissue culture were investigated. These techniques provide the theoretical support for breeding a large number of seedlings and promoting their applications in gardens. Looking forward to the prospects of the application of Cremastra appendiculata(D. Don) Makino in gardens, the aim is to promote its widespread utilization and provide references for trait research and genetic breeding, thereby achieving the coordinated development of medicinal and garden applications of Cremastra appendiculata(D. Don) Makino, enriching the variety of garden plants, improving the quality of landscapes, and ensuring the supply of medicinal resources.
Analysis of High Yield and Stability of New Maize Varieties in Guizhou Province Based on AMMI Model and GGE Biplot
YANG Tong-wen;DUAN Ming-yu;LI Hui;[Objective] In order to screen out varieties with high yield and stable yield, as well as test sites with high discrimination and good representativeness in regional trials in Guizhou Province.[Method] The AMMI model and GGE biplot were used to analyze the yield, stability, and adaptability of 7 maize varieties in Guizhou Province from 2021 to 2022 in 7 test sites, and to evaluate the discriminability and representativeness of the pilot projects.[Result] In 2021, Gengheyu 582, Bicheng 168, and Jinyu 631 had higher yield, while Jinbaiyu 6 and Gengheyu 582 had better stability; test sites in Liuzhi, Zhenfeng, and Liupanshui had better discriminability, while test site in Bijie, Liupanshui, and anshun had stronger representativeness. In 2022, Gengheyu 582 and Zunshi 2101 had higher yield, while Gengheyu 582, Jinyu 631, and Zunshi 2101 had better stability; test sites in Guiyang and Liuzhi had the better discriminability, while test site in Zhenfeng, Bijie had stronger representativeness.[Conlusion] Gengheyu 582 has good yield and stability, with the best overall performance; test site in Liuzhi had the best discriminability and test site in Bijie had the strongest representativeness.
Analysis of High and Stable Yield and Nitrogen and Phosphorus Utilization Efficiency of Luomai 49
GE Chang-bin;ZHU Lin-lin;QIN Su-yan;Luohe Agriculture and Rural Bureau;[Objective]In order to comprehensively understand the high yield stability, adaptability, disease resistance, and nitrogen and phosphorus utilization efficiency of Luomai 49.[Method]Combined with the data from the joint national wheat variety research and development of resistance in the southern part of the Huanghuai River from 2019 to 2020 and 2020 to 2021, as well as the production trials from 2020 to 2021, the high stability coefficient(HSC) and analysis of variance(ANOVA) methods were used to analyze the high yield stability, adaptability, disease resistance, quality, and nitrogen and phosphorus utilization efficiency of Luomai 49.[Result]The results showed that the yield of Luomai 49 was significantly higher than that of the control variety Zhoumai 18(P<0.01) in the national wheat variety joint research and development of resistance to Fusarium head blight and multi resistance groups in the southern part of the Huanghuai River region, as well as in the 2020-2021 production trial, demonstrating excellent high and stable yield; Luomai 49 demonstrated good adaptability in the joint national wheat variety research and development of resistance to Fusarium head blight and multi resistance groups in the southern part of the Huanghuai River in 2019-2020 and 2021-2021, as well as in the production trial in 2020-2021;Luomai 49 had good comprehensive disease resistance, including moderate resistance to Fusarium head blight, stripe rust, leaf rust, powdery mildew, and sheath blight.[Conclusion]Luomai 49 is a nitrogen and phosphorus high efficiency medium gluten wheat variety, which can be used for making traditional flour food such as Mantou and noodles.It can be promoted and applied as a green variety in the southern wheat region of the Huanghuai River.
Screening of Continuous Cropping Obstacle Resistance Chrysanthemum Varieties and Study on Its Resistance Mechanism
WANG Yan;JIANG Jing;MIAO Wei-hao;Based on the flower type, flower color and flower character, 55 cut chrysanthemum varieties were selected and the disease index were recorded to degrade the resistance level of cut chrysanthemum varieties, and then the resistance varieties were selected. The results indicated that the variety Nannong Libai showed high-resistance(HR) to continuous cropping obstacle, the varieties Nannong Bingjie and Nannong Yuanlv represent medium resistance(MR) to continuous cropping obstacle, and the varieties Menghong, Nannong Doulv and Nannong Daixue showed susceptible(S) to the continuous cropping obstacle, the remaining 49 cut chrysanthemum varieties were belonging to the high-sensitivity varieties(HS),accounting for 89.1% of the total tested varieties. The disease index of HS variety Nannong Fenying was 87.5%,which was 3.69 times than a HR variety Nannong Libai, the stem height of HS variety Nannong Libai was 95.00 cm, and the stem diameter was 5.34 cm, which was 1.44 and 1.22 times than that in the S variety Nannong Fenying. In addition, the leaf enzyme activities showed the positive correlation with varieties resistance level. The PPO,PAL and POD activities of plant leaves in HR variety Nannong Libai were 201.33,36.33 and 392.67 U/(g·min),respectively, which was 1.62,1.43 and 1.23 times than that in the HS variety Nannong Fenying. The activity of hydrogenase showed positively correlated with its variety resistance level. The soil enzyme activities showed as: HR > MR HS>S, the quantity of bacteria and fungi in the rhizosphere soil of HR varieties were 2.46 × 109 and 9.73×106 copies/g in the flowering time, which were 4.24 and 0.65 times higher than that in the HS variety Nannong Fenying soil.The study showed that the resistance mechanism of different cut chrysanthemum resistant varieties to continuous cropping obstacle was mainly by enriching beneficial microorganisms, maintain higher PAL and POD activities in leaf tissue, enhances soil enzyme activities, and then maintains the normal growth of roots and stems, and thus increasing the resists ability to the infection of pathogenic fungi. The result of this study is useful to tap and utilize the potential of chrysanthemum itself to provide an elite variety reserve for chrysanthemum disease resistance breeding in the future.












































































































































































































































































































































































































Research on Micro-climate and Medicinal Plant Growth Characteristics Under Different Canopy Densities in a Forest-Medicine Intercropping Model
MA Yi-xi;WANG Dong-li;LIU Jian-hua;[Objective] By monitoring the microclimate characteristics and the growth characteristics of medicinal plants under the forest medicine model, the relationship between microclimate change and plant growth characteristics was revealed. [Method] P. sylvestris forest in Zhanggutai was taken as the research object, and the stands with canopy densities of T0.2, T0.4 and T0.6 were constructed by thinning, and the forest medicine model was constructed by combining the planting of Radix Isatidis. [Result] The results showed that there was no significant difference in temperature and humidity between the forest floor and the ground surface with different canopy density, but the light intensity gradually weakened at noon and afternoon with the increase of canopy density, and T0.2 was significantly higher than T0.6(P<0.05). The temperature and light intensity in non-shaded areas were generally higher than those in shaded areas. The number of germination and emergence of T0.4 and the plant height in seedling stage were significantly higher than those in other forest lands with canopy density(P<0.05), and the plant height, biomass and yield in peak growth stage were significantly higher than T0.2(P<0.05). And the growth of plants in shaded areas was generally better than that in non-shaded areas. There is a positive correlation between microclimate indicators and plant growth indicators, while there is a negative correlation between plant growth and microclimate. [Conclusion] When planting Radix Isatis under P. sylvestris forest, a moderate canopy density should be established, and water management should be strengthened during the germination and emergence stage of plants in dryland areas without shade.
Teaching Reform of Cognition Practice in Marine Technology Specialty with the Goal of Improving Internship Teaching Quality

Through years of teaching practice in the Marine Technology major at Dalian Ocean University,and based on an analysis of the current status of course implementation,with the goal of“improving the quality of internship teaching”,curriculum reform and practice have been carried out in terms of course design,teaching organization and implementation,course teacher team construction,and course performance evaluation and verification,in order to promote the construction of the university's “Provincial First Class Major in Marine Technology”and achieve the goal of cultivating talents in the marine technology major.
Vertical Distribution Characteristics of Nitrogen and Phosphorus Contents in the Sediment of Ganqu Wetland and Correlation with Water Environment Factors
HE Wen-jia;WANG Zhuang-zhuang;LI Hong-chi;The physical and chemical factors of the wetland water environment and nitrogen and phosphorus contents in the sediment at different depths were measured and analyzed in 2024.Multivariate statistical analysis was applied to characterize the vertical distribution of nitrogen and phosphorus contents in the sediment at different depths in Ganqu Wetland and the correlation with the water environment factors.The results showed that as the depth of the 0-20 cm sediment in Ganqu wetland increased,the nitrate nitrogen content showed a decreasing trend,there was no significant difference in ammonia nitrogen and total nitrogen content,the total phosphorus content showed an increasing trend.There was a correlation between the nitrogen and phosphorus content in the sediment of Ganqu wetland and physical and chemical factors such as water redox potential,total nitrogen and total phosphorus.
Effect of Shading on Growth and Active Ingredients of Eleutherococcus senticosus
LI Xiang-quan;AI Zhi-qiang;LIU Ji-yun;[Objective]To explore the suitable light conditions for the growth and development of E.senticosus.[Method] Shading measures were taken to regulate the light intensity of E.senticosus,and the effects of shading on the growth and development of 2-year-old E.senticosus seedlings,as well as the accumulation of three active ingredients(eleutheroside E,syringin,isofraxidin) in roots and stems were studied.[Result]Moderate shading(transmittance of 38.29%-49.12%) could increase the height,ground diameter and leaf area of E.senticosus seedlings,and promoted the accumulation of aboveground biomass of E.senticosus.Shading had a significant effect on the syringin content in the stems of E.senticosus seedlings.The syringin content was the highest under moderate shading( transmittance 49.12%).Moderate shading was beneficial to the accumulation of syringin content in stems,but shading had no obvious effect on the syringin content in the roots.The effect of shading on the content of eleutheroside E in the root and stem was not obvious.Shading had a significant effect on the isofraxidin content in the roots and stems,with the increase of light transmittance,the change trend was opposite.However,under the condition of moderate shading( light transmittance 49.12%),the isofraxidin content was relatively high in both roots and stems.[Conclusion]Site selection and shading regulation should be paid attention to in understory planting and all-light planting of E.senticosus seedlings.Appropriate shading measures are beneficial for the growth of stems and leaves and the accumulation of active ingredients in the seedlings of E.senticosus,and are one of the key factors affecting the yield and quality improvement of E.senticosus.
Population Genetic Analysis of Eleutherococcus senticosus in Northeast China with Genotyping by Sequencing(GBS)
LI Xiang-quan;LIU Yun-wei;AI Zhi-qiang;[Objective] To effectively protect the genetic resources of Eleutherococcus senticosus germplasm.[Method]Using genotyping-by-sequencing(GBS) technology, single-nucleotide-polymorphism(SNP) site mining were identified in 120 E. senticosus individuals from four populations in Northeast China, and their genetic structure and genetic diversity were analyzed.[Result]By population evolutionary tree analysis, principal component analysis and structure analysis,combined with the geographical distribution characteristics of E. senticosus, four populations of E. senticosus were clustered into three different genetic groups.But the genetic boundaries between populations were not obvious,there were some degrees of gene exchange. Analysis of the 103 546 high-quality SNPs exposed that populations of E. senticosus in Northeast China had lower genetic diversity(Private=1 943.25,π=0.239,Ho=0.235 3,He=0.234 3),there were close genetic similarities and little genetic differences among different groups,only moderate differentiation exists between Jilin and Liaoning(Fst=0.053 2),indicating that the genetic basis of E. senticosus in Northeast China was relatively narrow,which may be due to its clonal reproductive characteristics.[Conclusion]The results not only provide molecular tools for genetic studies of E.senticosus, but also contribute to germplasm conservation and utilization in Northeast China.

The main chemical composition of Poria cocos are polysaccharide,triterpenes,fatty acids,sterols and so on. There are eleven polysaccharide compounds and forty-two triterpenoids. Poria cocos has a variety of pharmacological effects,two powerful active ingredients of Poria cocos are polysaccharides and triterpenes. Polysaccharides of Poria cocos have functions of antitumor activities,liver protective effect,acts as a diuretic,antisenility effects,anti-inflammatory,decrease blood fat,enhance immunity,sleep-inducing effect. Triterpenes of Poria cocos has function of antitumor activities,liver protective effect,antisenility effects,anti- inflammatory,enhancing immunity.

Research Advances in Extraction, Isolation, Purification and Content Determination of Methods Flavonoids

Research and Application Advances of Yeast Two-hybrid Technique
LI Xian-kun et al(College of Horticulture and Gardening of Hainan University,Danzhou,Hannan 571737)Yeast two-hybrid,which studies the interaction among proteins in yeast eukaryotic cells by using genetics method,is a very effective molecular biology technology.It is widely used in the fields such as proteomics,cellular signal transduction and functional genomics.And it has become one of important experiment means in the research field of molecular biology.And many valuable discoveries have been achieved using this system.With the development of molecular biology,based on yeast two-hybrid,many techniques such as yeast one-hybrid,reverse two-hybrid,three-hybrid,and extranuclear two-hybrid were set up to be applied in the research on the interaction between protein-protein,protein-DNA,protein-RNA,and protein-ligand.The research advances of yeast two-hybrid and its relevant technologies and application were summarized.It could be forecast that yeast two-hybrid and its derived techniques would play more and more important role in the researches of the functional genomics and proteomics.
