Abstract:

[Objective]Using high-performance liquid chromatography to determine the content of chlorogenic acid in Artemisia argyi leaves and optimizing its extraction process through orthogonal experiments.[Method]Orthogonal experimental design was used to compare the different influencing factors of ultrasound assisted ethanol extraction method, with the content of chlorogenic acid in Artemisia argyi leaves as the index, to investigate the effects of extraction time, extraction temperature, and ethanol concentration on the content of chlorogenic acid, an effective component in Artemisia argyi leaves.And detect the content of chlorogenic acid in Artemisia argyi leaves from 5 different regions available on the market.[Result]The optimal extraction process was ultrasound assisted ethanol extraction, and the optimal extraction conditions were ultrasound extraction for 45 min, temperature of 60 ℃,and ethanol concentration of 60%.Under these conditions, the content of chlorogenic acid extracted was the highest.The primary and secondary factors affecting extraction were: ultrasound temperature>ethanol concentration>extraction time.The content of chlorogenic acid in five different regions of Artemisia argyi leaves available on the market were different detected by this method.Among them, the chlorogenic acid content in Artemisia argyi leaves from Shandong was the lowest, at 0.196%;the content of chlorogenic acid in Artemisia argyi leaves in Guangxi region was the highest, at 0.756%.[Conclusion]The extraction method is simple and feasible, and has advantages of fast analysis speed, good repeatability, and high accuracy, making it suitable for the determination of chlorogenic acid content in Artemisia argyi leaves.
